summary

introduction and objectives
Tissue engineering gives the opportunity for preparing biodegradable scaffold seeded with cells for bladder reconstructions purposes.
aim of the study
Aim of the study was the evaluation of biodegradable scaffold seeded with cells used in bladder augmentation in the animal model.
material and methods
Cell culture: 3T3 mouse fibroblasts were used in the experiment. Cells were multiplied up to total amount of 108 cells. Cells were grown on DMEM supplemented with 10% of FBS (Fetal Bovine Serum) and antibiotics in 5% CO2 atmosphere and proper humidity. Cells were seeded on biodegradable PGA scaffold (polyglycolic acid) after 1 week of monolayer culture. Animals: Experimental group consisted of 6 rats. Through abdominal approach animals were partially cystectomized and cystostomy was created. Bladders were augmented with previously in vitro prepared flap. 3 animals were sacrificed after 1 week. Sonographic examination of the bladder was done in the 3 other rats after cystostomy removing at the second week after surgery. These rats were sacrificed after a month. There were two control groups. In the first group rats underwent partial cystectomy. Healthy animals were in the second group. H&E and immunohistochemical staining (against von willebrandt factor, CD31 and CD34) of the explanted bladder specimens was done.
results
There were no complications during postoperative period. Augmented bladders volume were comparable to normal rat bladder. Microscopic examination revealed PGA flap with growing 3T3 cells sutured to the muscle layer of the bladder. Angiogenesis process was seen within the flap. There was no inflammation in the bladder muscle.
conclusions
Tissue engineered patch is a useful material for urinary bladder augmentation.
